    M-grid: Using Ubiquitous Web Technologies to create a Computational Grid

    There are many potential users and uses for grid computing. However, the concept of sharing computing resources excites security concerns and, whilst being powerful and flexible, at least for novices, existing systems are complex to install and use. Together these represent a significant barrier to potential users who are interested to see what grid computing can do. This paper describes m-grid, a system for building a computational grid which can accept tasks from any user with access to a web browser and distribute them to almost any machine with access to the internet and manages to do this without the installation of additional software or interfering with existing security arrangements

    The Individual-Level Dynamics of Bounded Partisanship

    Over the past half century, scholars have utilized a variety of theoretical and methodological approaches to study the attachment or identification voters have with political parties. However, models of partisan (in)stability ignore its bounded character. Making use of Mixed Latent Markov Models, we measure the change and stability of individual-level West German partisan identification captured over a 24-year period via the German Socio-Economic Panel (GSEOP). Results suggest that distinctive subpopulations exist that follow different patterns of partisan stability. One party’s loss is not necessarily another party’s gain

    Mycobacterium ulcerans infection causes a neglected tropical disease known as Buruli ulcer that is now found in poor rural areas of West Africa in numbers that sometimes exceed those reported for another significant mycobacterial disease, leprosy, caused by M. leprae. Unique among mycobacterial diseases, M. ulcerans produces a plasmid-encoded toxin called mycolactone (ML), which is the principal virulence factor and destroys fat cells in subcutaneous tissue. Disease is typically first manifested by the appearance of a nodule that eventually ulcerates and the lesions may continue to spread over limbs or occasionally the trunk. The current standard treatment is 8 weeks of daily rifampin and injections of streptomycin (RS). The treatment kills bacilli and wounds gradually heal. Whether RS treatment actually stops mycolactone production before killing bacilli has been suggested by histopathological analyses of patient lesions. Using a mouse footpad model of M. ulcerans infection where the time of infection and development of lesions can be followed in a controlled manner before and after antibiotic treatment, we have evaluated the progress of infection by assessing bacterial numbers, mycolactone production, the immune response, and lesion histopathology at regular intervals after infection and after antibiotic therapy. We found that RS treatment rapidly reduced gross lesions, bacterial numbers, and ML production as assessed by cytotoxicity assays and mass spectrometric analysis. Histopathological analysis revealed that RS treatment maintained the association of the bacilli with (or within) host cells where they were destroyed whereas lack of treatment resulted in extracellular infection, destruction of host cells, and ultimately lesion ulceration. We propose that RS treatment promotes healing in the host by blocking mycolactone production, which favors the survival of host cells, and by killing M. ulcerans bacilli

    Pharmacologic management of Mycobacterium ulcerans infection

    Introduction: Pharmacological treatment of Buruli ulcer (Mycobacterium ulcerans infection; BU) is highly effective, as shown in two randomized trials in Africa. Areas covered: We review BU drug treatment–in vitro, in vivo and clinical trials (PubMed: ‘(Buruli OR (Mycobacterium AND ulcerans)) AND (treatment OR therapy).’ We also highlight the pathogenesis of M. ulcerans infection that is dominated by mycolactone, a secreted exotoxin, that causes skin and soft tissue necrosis, and impaired immune response and tissue repair. Healing is slow, due to the delayed wash-out of mycolactone. An array of repurposed tuberculosis and leprosy drugs appears effective in vitro and in animal models. In clinical trials and observational studies, only rifamycins (notably, rifampicin), macrolides (notably, clarithromycin), aminoglycosides (notably, streptomycin) and fluoroquinolones (notably, moxifloxacin, and ciprofloxacin) have been tested. Expert opinion: A combination of rifampicin and clarithromycin is highly effective but lesions still take a long time to heal. Novel drugs like telacebec have the potential to reduce treatment duration but this drug may remain unaffordable in low-resourced settings. Research should address ulcer treatment in general; essays to measure mycolactone over time hold promise to use as a readout for studies to compare drug treatment schedules for larger lesions of Buruli ulcer

    Star clusters in the Sh2-132 complex: clues about the connection between embedded and open clusters

    Embedded clusters are formed in molecular clouds where massive stars can produce HII regions. The detailed embedded-open cluster evolutionary connection as well as the origin of associations are yet to be unveiled. There appears to be a high infant mortality rate among embedded clusters and the few survivors evolve to open clusters. We study the colour-magnitude diagrams and structure of the star clusters related to the Sh2-132 HII region using the 2MASS database. Cluster fundamental and structural parameters are determined via MS and PMS isochrones and stellar radial density profiles. We report the discovery of four clusters. One of them is projected a few diameters away from the optical cluster Teutsch\,127 and appears to be deeply embedded, seen only in the infrared. Evidence is found that we are witnessing the dynamical transition from an embedded to an open cluster. An additional cluster is also close to Teutsch\,127 and might be associated with a bow-shock. We also study the CMD and structure of the open cluster Berkeley\,94 in Sh2-132 and a new cluster which is projected in the outskirts of the complex. Finally, we searched for star clusters around the two known Wolf-Rayet stars in the complex. One of them appears to be related to a compact cluster.     Is it all about storytelling? Living and learning hereditary cancer on Twitter

    Storytelling has long been used as a theoretical framework for understanding how we share information and learn about health – and illness – online. But is it all about storytelling on social media platforms? To explore how and to what extent personal stories shape health content on these platforms, the article presents an analysis of tweets discussing the BRCA gene mutation – a hereditary cancer condition. Theoretically, the study advances a new conceptual framework to explore social media practices within issue–based and long–lived social media threads. Methodologically, it develops a qualitative, platform–oriented discourse analytic approach. Findings show that non narrative content is actually more common than storytelling in Twitter conversations about BRCA, with a number of patient advocates acting as gatekeepers of scientific information. Most BRCA storytelling is mediated and shared in third person, with those at the heart of these stories becoming exemplars within the BRCA ‘subculture’

    Thinking about politics

    There are distinctive modes of thinking about politics, three of which are discussed here. A mode consists of a characteristic domain of relevance, filing system, and grammar of beliefs. A person relying on Mode A treats politics as an extension of interpersonal experience. A person relying on Mode B organizes political thinking around a set of salient group identifications. A person relying on Mode C views public objects in terms of their consequences for collective goods. The three modes are illustrated by applying them to concrete issues in a hypothetical manner: Vietnam, bussing, and attitudes toward presidential candidates.     Adaptive Management of Bull Trout Populations in the Lemhi Basin

    The bull trout Salvelinus confluentus, a stream-living salmonid distributed in drainages of the northwestern United States, is listed as threatened under the Endangered Species Act because of rangewide declines. One proposed recovery action is the reconnection of tributaries in the Lemhi Basin. Past water use policies in this core area disconnected headwater spawning sites from downstream habitat and have led to the loss of migratory life history forms. We developed an adaptive management framework to analyze which types of streams should be prioritized for reconnection under a proposed Habitat Conservation Plan. We developed a Stochastic Dynamic Program that identified optimal policies over time under four different assumptions about the nature of the migratory behavior and the effects of brook trout Salvelinus fontinalis on subpopulations of bull trout. In general, given the current state of the system and the uncertainties about the dynamics, the optimal policy would be to connect streams that are currently occupied by bull trout. We also estimated the value of information as the difference between absolute certainty about which of our four assumptions were correct, and a model averaged optimization assuming no knowledge. Overall there is little to be gained by learning about the dynamics of the system in its current state, although in other parts of the state space reducing uncertainties about the system would be very valuable. We also conducted a sensitivity analysis; the optimal decision at the current state does not change even when parameter values are changed up to 75% of the baseline values. Overall, the exercise demonstrates that it is possible to apply adaptive management principles to threatened and endangered species, but logistical and data availability constraints make detailed analyses difficult
